I would suggest exiting and restarting PP.
But otherwise, I see a whole lot of suspicious things in your screenshot (separators in the toolbar, columns in your outbox) which could be indicative of some GUI issues. If you still have the problem after restarting PP, you may need to send me your ini settings so that I can try to duplicate it. What version of POP Peeper are you using (main menu: Help / About)?

As for the columns in the outbox -- in your screenshot, only the "account" column was visible, which doesn't seem very useful. In your last screenshot, "subject" is now visible although the "account" column width is probably larger than it should be. If you right-click on the column header, you can then select "reset all column sizes" while viewing the "outbox"; unless you really want it that way (I also realize you probably only have the "outbox" page selected so as to hide your inbox messages).
Ok, as to your main question -- POP Peeper looks for a file named poppeeper.ini in 2 locations (in this order):
1) The current folder where you're running PP (ie. F:\POPPEEPER\POPPeeperPro-v453\)
2) The AppData folder
So it sounds like you want a fresh start on the portable drive -- no accounts, default settings and no messages. Here's how:
1) Exit POP Peeper -- make sure that no instance of POP Peeper is currently running
2) Open a command prompt
3) type (followed by <enter> after each line):
f:
cd POPPEEPER\POPPeeperPro-v453\
4) type (and then press <enter>):
poppeeper -ini
5) This will bring up the ini manager, press the "new" button
-> by default, it should specify a "poppeeper.ini" file in the f:\poppeeper\...\ folder -- this is what you want so press "save"
6) Go through the setup and make sure you select the "store data for portability" option on the 2nd page (it should be selected by default)
You won't need to use the command prompt again, that's just necessary the first time to create the ini file. From now on, when you run poppeeper from your usb drive on F:, it will automatically find and use the poppeeper.ini file on your usb drive. Likewise, if you run POP Peeper on your old C: drive, it will use the original poppeeper.ini that's in your AppData folder.

That's the system.ini file located under POP Peeper\Plugins\Profile\ which is used for the Profile Picture plugin. It is not an ini file that is usable for loading your accounts/settings.
