What is the specific error that is reported? Gmail sometimes gives a hint in the error.
Otherwise, yes, I would suggest just making another app-specific password. Google won't allow you to see old app-passwords, but I would recommend deleting the old one.

If you have POP Peeper Pro, there's a rule in POP Peeper that may be causing this. Originally, the rule would hide and mark read any message you sent to yourself, but this was later updated so that messages that you to/cc yourself would not be affected (it would only be affected if you BCC'd yoursel...

You need to make sure that you've provided POP Peeper with the app-specific password. I'm fairly certain that Gmail only responds with "Application-specific password required" if it is still trying to login with your main password.

The only maintenance that POP Peeper runs is compacting the databases, and that's only performed every 90 days by default (Tools / Options / Storage). Once you enable 2FA on your account and then create an app-password for POP Peeper to use, that password will behave just like a normal password; ie. you don't have to 2FA everytime to check mail on POP Peeper. PP starts as minimized by default when it's run at startup, but shouldn't when you run it manually. So if you start it manually and it crashes, try to notice if the main window appeared.
